Its high selectivity allows for one-step purification. Affinity gel can be used in either denaturing or non-denaturing conditions. IDA or NTA? Briefly, NTA chelates metal ion tighter that IDA and metal leakage low, but difficult to elute the target proteins. IDA chelates metal ion loosely and metal leakage is more but easier to elute the target proteins.
